RESUMO

Angaracris rhodopa (Fischer et Walheim), Calliptamus abbreviatus (Ikonnikov), Myrmeleotettix palpalis (Zubowsky), and Oedaleus decorus asiaticus (Bey-Bienko) are the main grasshoppers that harm the natural grassland in the Hexi Corridor in Gansu, northwest China. In this study, the MaxEnt model was employed to identify the key environmental factors affecting the distribution of the four grasshoppers' habitats and to assess their distribution under current and future climate conditions. The aim was to provide a basis for grasshopper monitoring, prediction, and precise control. In this study, distribution of suitable habitats for A. rhodopa, C. abbreviates, M. palpalis, O. decorus asiaticus were predicted under current and future climatic scenarios using the Maxent model. The average AUC (area under the ROC curve) and TSS (true skill statistic) values of the four grasshoppers were greater than 0.9, and the simulation results were excellent and highly reliable. The mean annual precipitation was the main factor limiting the current range of suitable areas for these four species. Under the current climate, A. rhodopa, C. abbreviatus, and O. decorus asiaticus were mainly distributed in the central and eastern parts of the Hexi Corridor, and M. palpalis was distributed throughout the Hexi Corridor, with a suitable area of 1.29 × 104, 1.43 × 104, 1.44 × 104, and 2.12 × 104 km2, accounting for 13.7%, 15.2%, 15.3%, and 22.5% of the total area of the grasslands in the Hexi Corridor, respectively. The highly suitable areas of A. rhodopa, C. abbreviatus, and O. decorus asiaticus were mainly distributed in the eastern-central part of Zhangye City, the western part of Wuwei City, and the western and southern parts of Jinchang City, with areas of 0.20 × 104, 0.29 × 104, and 0.35 × 104 km2, accounting for 2.2%, 3%, and 3.7% of the grassland area, respectively. The high habitat of M. palpalis was mainly distributed in the southeast of Jiuquan City, the west, middle, and east of Zhangye City, the west of Wuwei City, and the west and south of Jinchang City, with an area of 0.32 × 104 km2, accounting for 3.4% of the grassland area. In the 2030s, the range of A. rhodopa, C. abbreviatus, and O. decorus asiaticus was predicted to increase; the range of M. palpalis will decrease. The results of this study could provide a theoretical basis for the precise monitoring and control of key areas of grasshoppers in the Hexi Corridor.

Natural grasslands are being progressively degraded around the world due to climate change and socioeconomic factors. Most of the drivers, processes, and consequences of grassland degradation are studied separately, and it is not yet clear whether the change characteristics and influence factors of adjacent areas of grassland are identical. We analyzed changes in grassland area and quality, and the influences of climate changes and socioeconomic factors from 1980-2018 in Maqu County, Xiahe County and Luqu County on the eastern Qinghai-Tibet Plateau (QTP). We found that areas with high and medium coverage grassland in Maqu County and Luqu County decreased continuously with time, while low coverage grassland areas increased in three counties. In Xiahe County, the medium coverage grassland area reduced with time (except for 2010), while the high and low coverage grassland areas increased. The actual net primary productivity of the three counties showed a downward trend. In Maqu County, the total grassland area had an extremely significant positive correlation with number of livestock going to market, commodity rate, gross domestic product (GDP), primary industry, tertiary industry, household density, and levels of junior middle school education and university education in the area. In Luqu County, the total grassland area high coverage grassland area were significantly negatively correlated with total number of livestock, secondary industry, levels of primary school education, and temperature. Ecological education was positively correlated with high coverage grassland, and negatively correlated with low coverage grassland in all three areas. The results of this study suggest that the best ways to restore the area and quality of grasslands in these areas would be to reduce the local cultivated land area and slow down the development of the primary and tertiary industries in Maqu County, and to control industry development and the total number of livestock in Luqu County. This study also suggests that improving education level and strengthening the level of ecological education are conducive to the restoration of grasslands.

Ligularia virgaurea and Ligularia sagitta are two species of poisonous plants with strong invasiveness in natural grasslands in China that have caused considerable harm to animal husbandry and the ecological environment. However, little is known about their suitable habitats and the key environmental factors affecting their distribution. Although some studies have reported the distributions of poisonous plants on the Qinghai-Tibet Plateau (QTP) and predicted their potential distributions at local scales in some regions under climate change, there have been few studies on the widespread distributions of L. virgaurea and L. sagitta. In this study, we recorded 276 and 118 occurrence points of L. virgaurea and L. sagitta on the QTP using GPS, and then used the MaxEnt model to predict the distribution of suitable habitats. Results showed that (1) under current climate conditions, L. virgaurea and L. sagitta are mainly distributed in southern Gansu, eastern Qinghai, northwestern Sichuan, eastern Tibet, and southwestern Yunnan, accounting for approximately 34.9% and 39.8% of the total area of the QTP, respectively; (2) the main environmental variables affecting the distribution of suitable habitats for L. virgaurea and L. sagitta are the Human Footprint Index (52.8%, 42.2%), elevation (11%, 4.4%), soil total nitrogen (18.9%, 4.2%), and precipitation seasonality (5.1%, 7.3%); and (3) in the future, in the 2050s and 2070s, the area of habitat of intermediate suitability for L. virgaurea will spread considerably in northwest Sichuan, while that of high suitability for L. sagitta will spread to eastern Tibet and western Sichuan.

Exploring the activity patterns of small mammals is important for understanding the survival strategies of these animals, such as foraging and mating. The purpose of the present study was to determine the activity of free-living plateau pikas (Ochotona curzoniae) in different months and seasons (cold and warm seasons), with a particular emphasis on the effects of weather condition. Based on a camera-trapping survey conducted from October 2017 to September 2018, we evaluated the activity patterns and activity levels of plateau pikas inhabiting the eastern Qinghai-Tibet Plateau in China. The effects of environmental factors on the activity of plateau pikas were examined using the generalized additive mixed model (GAMM). The results showed that: (1) The plateau pikas exhibited unimodal patterns of activity during the cold season (October-April). During the warm season (May-September), the activity patterns of the plateau pikas were bimodal. Their activity levels were highest in June. (2) During the cold season, their activity levels rose gradually over the course of the day to a peak near noon, and they were not significantly higher after sunrise than they were before sunset. During the warm season, their activity peaks were in the morning and afternoon, and their activity levels were substantially lower after sunrise than they were before sunset. (3) The plateau pikas were more active under conditions with lower ambient temperatures and precipitation during the cold and warm seasons. While relative air humidity was positively correlated with the activity of the plateau pikas during the warm season, wind speed was negatively correlated with the pikas' activity during the cold season. Overall, these results collectively indicate that plateau pikas occupy habitats with cool and less windy microclimates during the cold season, and with cool and moist microclimates during the warm season. Information on the time allocation of pikas' activity levels during different seasons should provide a baseline for understanding their potential for adaptation to climate change.

Acoustic communication plays a vital role in passing or sharing information between individuals. Identifying the biological meaning of vocal signals is crucial in understanding the survival strategies of animals. However, there are many challenges in identifying the true meaning of such signals. The plateau pika (Ochotona curzoniae) is a call-producing mammal endemic to the Qinghai-Tibet plateau (QTP) and considered a keystone species owing to its multiple benefits in alpine rangeland ecosystems. Previous studies have shown that plateau pikas emit alarm calls as part of their daily activities. However, only field observations have been used to identify these alarm calls of the plateau pika, with no attempts at using playback experiments. Here, we report the alarm calling of plateau pikas through field observations as well as simulated predator and playback experiments in the Eastern QTP from 2021 to 2022. We found that both female and male adults emitted alarm calls, the signals of which comprised only one syllable, with a duration of 0.1-0.3 s. There were no differences in the characteristics between the observed alarm calls and those made in response to the simulated predator. The duration of the alarm call response varied with altitude, with plateau pikas living at higher altitudes responding at shorter durations than those at lower altitudes.

Endocrine cells can secrete a variety of gastrointestinal hormones that regulate gastrointestinal digestion and absorption, which, in turn, play an important role in animal growth, metabolism, and acclimation. The small alpine mammals, plateau zokor (Eospalax baileyi) and plateau pika (Ochotona curzoniae), live in a unique ecotope with cold, hypoxic environments and short plant-growing seasons, resulting in differential adaptive digestive strategies for foods. Studying the distribution pattern of endocrine cells in the gastrointestinal tract (GIT) of these two animals can lead to a better understanding of the survival strategies of animals in an alpine environment. In this study, we used histochemical and immunohistochemical methods to compare the distribution pattern of argyrophilic cells and the expression of 5-HT cells, Gas cells, and Glu cells in the GIT of the plateau zokor with those of the plateau pika. The results showed that these endocrine cells we studied were widely distributed in the gastrointestinal organs of both these small mammals, and their morphology and distribution location in the GIT were almost the same. However, there were significant differences in the distribution density of argyrophilic cells between different organs in the GIT. The distribution density of argyrophilic cells in the duodenum, jejunum, ileum, and rectum of plateau zokor was significantly lower than that of plateau pika (p < 0.05) and, in the cecum of plateau zokor, was significantly higher than that of plateau pika (p < 0.001). The positive expression of 5-HT cells in the corpus I, corpus II, and pylorus of the stomach, duodenum, ileum, and rectum of plateau zokor was significantly higher than that of plateau pika (p < 0.01). In addition, the positive expression of Glu cells in the cecum was significantly higher (p < 0.01) and in the duodenum and colon was significantly lower (p < 0.05) in the plateau zokor than in the plateau pika. We conclude that the distribution pattern of endocrine cells in the GIT is consistent with the respective animals' diets, with the plateau zokor feeding on high-fiber roots and plateau pika preferring to intake the aboveground parts of plants with lower fibers.

There are considerable challenges involved in studying the behavior of subterranean rodents owing to the underground nature of their ecotope. Seismic communication plays a crucial role in the behavior of subterranean rodents, particularly solitary ones. The plateau zokor (Eospalax baileyi), a solitary subterranean rodent species endemic to the Qinghai−Tibet Plateau, will usually occupy empty neighboring tunnels in order to extend their territory. Little is known, however, about the process of territorial occupation or the function of animal communication when occupation is taking place. Based on previous studies of subterranean rodent communication, we hypothesized that plateau zokors use seismic signals to detect neighboring tunnels and then occupy them when it was found their neighbors were absent. To test this, we placed artificial tunnels close to active original zokor tunnels to simulate the availability of an empty neighboring tunnel, and then the seismic signals when a zokor chose to occupy the empty artificial tunnel were recorded. The results showed that the frequency of zokors occupying artificial empty tunnels within 48 h was 7/8, In all of these instances, the zokors generated seismic signals before and after occupation of the empty artificial tunnel. The number of seismic signals generated by the zokors increased significantly (p = 0.024) when they detected and occupied the artificial tunnels, compared to those generated in their original tunnels without the presence of an artificial tunnel alongside. Inside the original tunnels, the inter-pulse time interval of the seismic signals was significantly higher (p < 0.001), the peak frequency of these signals was significantly higher (p < 0.01), and the energy of the signals was significantly lower (p = 0.006), compared with those when an artificial tunnel was positioned next to the original. The results of this study suggest that plateau zokors first generate seismic signals to detect empty neighboring tunnels and that they are empty. In the absence of neighbor plateau zokors, they occupy the empty tunnels to extend their own territory.

The morphological and histological traits of the gastrointestinal tract (GIT) enable the animal to perform some specific functions that enhance the species' adaptability to environments. The plateau zokor (Eospalax baileyi) is a subterranean rodent that mainly forages on plant roots in the Qinghai-Tibet Plateau, but little is known about the mechanism by which the plateau zokor digests roots that have high fiber contents. In this study, we used comparative anatomy methods to compare the morphological and histological traits of the GIT of both the plateau zokor and the plateau pika (Ochotona curzoniae), a small, fossorial lagomorph that forages aboveground plant parts, in order to clarify the traits of the plateau zokor's GIT and to understand its adaptations to high-fiber foods. The results showed that the foods which plateau zokors eat have a higher fiber content than those which the plateau pikas eat. The plateau zokor has a double-chambered and hemi-glandular stomach (the tubular glands are only in the gastric corpus II, and the gastric fundus is keratinized), whereas the plateau pika has a simple, wholly glandular stomach. The gross morphological indicators (organ index and relative length) of the GIT were significantly lower in the plateau zokor than they were in the plateau pika (p < 0.001). However, the thickness of the gastric corpus II mucosal layer and the gastric fundus muscle layer are significantly higher in the plateau zokor than they are in the plateau pika (p < 0.001), and the thickness of each layer of intestinal tissue is higher in the plateau zokor than it is in the plateau pika. Additionally, the small intestinal villi also are higher and wider in the plateau zokor than they are in the plateau pika. Our results suggest that instead of adapting to digest the high-fiber diet by expanding the size of the GIT, the plateau zokor has evolved a complex stomach and a well-developed gastrointestinal histological structure, and that these specialized GIT structures are consistent with an optimal energy-economy evolutionary adaptation strategy.

Defensive behaviors are a response to immediate and potential threats in the environment, including abiotic and biotic threats. Subterranean rodents exhibit morphological and physiological adaptions for life underground, and they will seal with mounds and additional plugs when their burrow opened. However, little is known about the factors driving this defensive behavior. In this study, we selected a subterranean rodent, plateau zokor (Myospalax fontanieri), as a species to investigate (both in the laboratory and in the field) the possible factors responsible for burrow-sealing behavior. Our results showed that: (1) In the laboratory, the burrow-sealing frequency of plateau zokor in response to five factors were as follows: oxygen (52.63%) > light (34.58%) > temperature (20.24%) > gas flow (6.48%) > sound/control (0%). Except for light, the burrow-sealing frequency in response to other factors was significantly lower than that in response to oxygen (P < 0.05). (2) Burrow-sealing behavior in response to each treatment did not differ significantly between males and females in the laboratory experiment. (3) In the field, during the animal's active periods in both the cold and warm season, the burrow-sealing frequency under the oxygen treatment was higher than that under the light and temperature treatments. Plateau zokors were found not to be sensitive to these treatments during their inactive periods during both the cold and warm season. (4) The latency to reseal the burrow showed no obvious differences between each treatment both in the laboratory and in the field. In conclusion, the main factor that influences the burrow-sealing behavior of plateau zokors is the variation in oxygen concentration, and this defensive behavior is related to their activity rhythm.

BACKGROUND: Plateau zokor (Myospalax baileyi) is a subterranean rodent endemic to the Tibetan Plateau. This species has been generally viewed as a pest in China due to the competition for food with livestock and also causing soil erosion. As a result, plateau zokor has been the target of widespread poisoning or trapping campaigns designed to control or eliminate it since 1970s. But there is little research on the effect of plateau zokor on plant diversity in alpine rangelands. Therefore, objectively evaluating the positive effects of the plateau zokors disturbance on their living environment and plant communities is of great significance to understand the function of plateau zokor in alpine ecosystem. METHODS: Here, we selected three rangelands (alpine meadow, alpine steppe and alpine shrub meadow) in which plateau zokors are typically distributed on the Tibetan Plateau, and five zokor mound density gradients were selected in each rangeland type to study the effects of the mounds on soil moisture and temperature related to plant species diversity. RESULTS: The results showed that, with the mound density increasing, the soil temperature decreased significantly in all three rangeland types, and the soil moisture significantly increased in all three rangeland types. In the alpine meadow, both the plant diversity and cumulative species richness increased significantly with increasing mound density. The increase in broad-leaved forbs is the main reason for the increase of plant diversity in the alpine meadow disturbed by zokor mounds. In the alpine steppe, the plant diversity decreased significantly with increasing mound density, while the cumulative species richness initially decreased and then increased. In the alpine shrub meadow, the plant diversity first increased and then decreased with increasing mound density as did the cumulative species richness. In conclusion, plateau zokor mounds dominated the distribution of soil moisture and temperature and significantly affected plant diversity in these three rangelands on Tibetan Plateau; the results further deepen our understanding toward a co-evolved process.

Plants are particularly sensitive to climate change in alpine ecosystem of the Tibetan Plateau. The various mountain micro-climates provide a natural gradient for space-for-time substitution research that plant responses to climate change. In this study, we surveyed the plant community in term of species composition, diversity and biomass across 189 sites on a hill of the Tibetan Plateau and analysed the individual and integrated effects of soil temperature and moisture on the plant community. The results showed that, at the quadrat scale, there were decrease in richness of 1.08 species for every 1 °C increase in soil temperature and 3.56 species for every 10% decrease in soil moisture. The integrated effects of increasing soil temperature and decreasing moisture are expected to lead to a rapid decrease in species richness. Biomass had no significant correlation with soil temperature but significantly decreased with soil moisture decreasing (p < 0.01). Biomass would decrease when soil moisture was below 20%, no matter how the change of soil temperature. We also found that gramineae and perennial forbs were sensitive to climate change. With soil temperature increased, the proportion of gramineae increased, whereas the proportion of perennial forbs decreased. The integrated effects of soil temperature increasing and moisture decreasing caused a shift from sedge-controlled to gramineae-controlled communities in alpine meadow. This study not only enhances our understanding of mountain plant community dynamics under climate change, but also predicts the shift of vegetation response to climate change on high-elevation alpine meadow.

In this study, we use three mitochondrial markers, cytochrome b gene (Cyt b), NADH dehydrogenase subunit 4 (ND4) and control region (D-loop) to investigate the phylogenetic relationships of extant zokor species in Mysopalacinae. The phylogenetic tree constructed based on Cyt b strongly supports the monophyly genera Eospalax and Myospalax with E. fontanierii being the most ancient species in Eospalax. Further phylogenetic analyses of four species of Eospalax based on ND4 and D-loop sequences revealed two clades that correspond to two geographical distributions. The basal clade includes E. cansus which is mainly found on Loess Plateau (LP) and another clade including E. baileyi, E. smithii and E. rufescens that inhabits areas above 2000 m on Qinghai-Tibetan Plateau (QTP) and Qinling Mountains. Geographical events of QTP and LP may have played a major role in the diversification and evolution of Mysopalacinae.

Mysopalacinae (zokors) is a group of fossorial rodents for which the taxonomy has yet to reach consensus. Furthermore, due to their fossorial lifestyle, little is known about their ecology. Molecular data are important to elucidate such aspects. In this paper, the complete mitochondrial DNA genome of Gansu zokor (Eospalax cansus) of the type found in Lintan, China was determined. The genome is 16,354 bp in length and consists of 13 protein-coding genes, 22 tRNA genes, two ribosomal RNA genes, and two main non-coding regions (the control region and the origin of the light strand replication), the gene composition and order of which are similar to most other mammals. The overall base composition is T 30.0%, C 24.2%, A 33.5%, and G 12.3%, with an A + T bias of 63.5%. These mitogenome sequence data are potentially important for evolutionary, population genetic, and ecological studies of the Mysopalacinae.

Vanillin is a food flavoring agent widely utilized in foods, beverages, drugs, and perfumes and has been demonstrated to exhibit multiple pharmacological activities. Given the importance of glucuronidation in the metabolism of vanillin, the UDP-glucuronosyltransferase conjugation pathway of vanillin was investigated in this study. Vanillin glucuronide was identified by high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) and a hydrolysis reaction catalyzed by ß-glucuronidase. The kinetic study showed that vanillin glucuronidation by HLMs and HIMs followed Michaelis-Menten kinetics and the kinetic parameters were as follows: 134.9 ± 13.5 µM and 81.3 ± 11.3 µM for K(m) of HLMs and HIMs, 63.8 ± 2.0 nmol/min/mg pro and 13.4 ±2.0 nmol/min/mg pro for Vmax of HLMs and HIMs. All UDP-glucuronosyltransferase (UGT) isoforms except UGT1A4, 1A9, and 2B7 showed the capability to glucuronidate vanillin, and UGT1A6 exerted the higher V(max)/K(m) values than other UGT isoforms for the glucuronidation of vanillin when assuming expression of isoforms is similar in recombinant UGTs. Kinetic analysis using liver microsomes from six studied speices indicated that vanillin had highest affinity for the monkey liver microsomes enzyme (K(m) = 25.6 ± 3.2 µM) and the lowest affinity for the mice liver microsomes enzyme (K(m) = 149.1 ± 18.4 µM), and intrinsic clearance was in the following order: monkey > dog > minipig > mice > rat ~ human. These data collectively provided important information for understanding glucuronidation of vanillin.
